When I test Cisco Router and TFTPd64 by changing security as High and Read only settings in server, the error show as (Permission denied)

I would think you are correct, if the TFTP server is not started, it will never able to open the socket itself. I would think the answer needs to be: Configure the TFTP server allow writing towards the specified directory.
