2001:db8:1234 uses up 48 bits (as one hexadecimal digit is equivalent to 4 bits (2^4 = 16 possibilities). 64 bits are used for the hosts so 16 bits remain. 2^7 = 128 and sufficient to provide for 100 locations. 2^9 = 512 and sufficient to provide for 300 subnets.

The 7 bit for locations is in 2001:db8:1234 The 9 bit for subnet is in 16bit following 2001:db8:1234 The 64 bit for hosts, this quetion doesn't refer to, is in 64 bit following 16bit.
